Subject: Binding affinities--PGE2/PGF2a
According to radioreceptor binding studies analyzed by Scatchard analysis in our lab (Milo Wiltbank's), in the bovine mid-cycle corpora lutea plasma membrane:
PGF2a Kd = 7.6 - 9.2 nM
PGE1 Kd = 11 nM
From inhibition curves, the IC50s for 4 prostaglandins also in the bovine mid-cycle corpus luteum are as follows:
PGD2--22 nM
PGE1--14 nM
PGE2 --8 nM
PGF2a--22 nM
Be aware that both PGD2 and PGE2 appear to cross-react with the PGF2a receptor (FP), but PGE1 does not. Using either PGD2 or PGE2 in a tissue that contains high levels of FP receptor, such as the CL, will confound the results since the FP receptor may be activated. We have little evidence for a PGD2 receptor (DP). Preliminary experiments show that a specific DP receptor antagonist (BW245-C) does not inhibit radioactive PGD2 binding in the bovine CL.
