Flow Cytometry Services

The Division of Cardiovascular Medicine provides a flow cytometry service to other divisions both within and outside of the School of Clinical Sciences.

Brief Description

The division has two state of the art Becton Dickinson machines:

  • Becton Dickinson LSR11 fitted with blue, red, and UV lasers for excitation of fluorchromes at 488, 633 and 325nm respectively.
  • Becton Dickinson FacsCanto II with both blue and red lasers for excitation at 488 and 633nm. This machine also has a HTS unit for processing of 96 well plates rather than FACS tubes.  

The analytical capabilities of these flow cytometers are second to none within the University. Both machines are set up with FACSDiva software, but this is compatible with other flow cytometry software and it is possible to download fcs files for analysis in WinMDI, FlowJo etc.

The machines are analytical machines and do not have cell sorting capabilities. If you want to sort, please contact Nina Lane in the Division of Immunology for this service.

  • Cell surface antigens
  • Internal antigens using permeabilised cells
  • Detection of FITC, PE, PE-Cy7, Texas red, PerCP, PerCP-Cy5, PE-Cy5, PI, GFP, DAPI, Indo, Alexa 350, APC, APC-Cy7, Alexa 647, Hoechst 33342
  • Intracellular Calcium studies eg. using Fluo and Indo dyes
  • Apoptosis assays/microparticle formation and detection
  • Cytometric bead assays

Our machines will run many of the *bead assay technologies including Luminex, Bender microsystems, Merck BeadPlex as well as BD CytometricBeadArrays.

*bead assays - they may be described as an ELISA with antibody bound to a bead instead of a plate. The advantage is that you can run many together at the same time on the same sample and therefore build up an array. Only a small amount of sample is needed. 

General Information

All work must be performed according to laboratory safety rules. The Safety officer for Cardiovascular Medicine is Jane May.

The service is available weekdays, between 9:00am and 5.30pm.

Charges

There is no charge for initial discussion, machine setup and template preparation.

One hour (supervised) = £80

One hour (trained user; unsupervised) = £40

The above costs include machine use and FACsFlow diluent where needed, but no other consumables

FACS Tubes can be provided at a cost of £15.50 per pack of 500, otherwise compatible tubes suitable for these Becton Dickinson machines must be used at all times.

These tubes include BD Falcon, cat.352052 or Sarstedt cat.55.1579, (they are 5ml, round bottom, PS, 12 x 75mm)
