Investigative Laboratories in Cell Biology Using a Host-Parasitoid Model
A series of exercises were developed in the cell biology laboratory using a host-parasite system: the tobacco hornworm and an inoffensive parasitoid wasp. Exercises are fully investigative and no initial schedule is given to the students who, after becoming acquainted with the system, raise questions and decide as a group what they wish to study next (Cell morphology? Cytoskeleton? Protein synthesis?). This workshop involved introduction to the system of study, phase contrast microscopy, and dig

Transposon Mutagenesis of Rhodobacter sphaeroides
Creation and characterization of mutants is the basis for any genetic analysis. This exercise demonstrates a simple, safe procedure for transposon mutagenesis of Rhodobacter sphaeroides, a purple non-sulfur photosynthetic bacteria. Students perform the mutagenesis by mating a transposon-carrying plasmid from Escherichia coli to R. sphaeroides and then selecting for the drug resistance carried on the transposon. Only R. sphaeroides carrying the transposon in the chromosome survive the selection.
