Abstract:
Prevalent diseases such as cancer and neurodegenerative disease are etiologically and pathologically complex. Strategies for successfully treating such conditions are expected to include combination therapies that can simultaneously target multiple disease-relevant pathways; however, identifying combination therapies de novo presents combinatorial challenges. The availability of large combination screening data sets in human cell lines has spurred the development of computational methods for predicting in vitro drug efficacies and synergies for complex diseases. Here I will present statistical models for uncovering both effective combination therapeutics and disease mechanisms using phenotypic screens and publicly available drug-target information. I will discuss how screening strategies can be improved by effectively incorporating interpretable computational models.
